Durobac tablets, which contain sulfamethoxazole and trimethoprim, can cause a range of side effects. Common side effects include headache, nausea, diarrhoea, and rash[5]. Less frequent or rare side effects involve:

  • Central nervous system effects: apathy, ataxia (loss of muscle control), convulsions, depression, fatigue, hallucinations, insomnia, nervousness, peripheral neuritis (nerve pain), vertigo, and weakness[1].
  • Ears and eyes: tinnitus (ringing in the ears), short periods of vision disturbance, and eye inflammation[5].
  • Hematological (blood): agranulocytosis, aplastic anemia, eosinophilia, hemolytic anemia, hypoprothrombinemia, leukopenia, megaloblastic anemia, methemoglobinemia, neutropenia, thrombocytopenia[1].
  • Hypersensitivity and severe allergic reactions: Stevens-Johnson syndrome, toxic epidermal necrolysis, allergic myocarditis, anaphylaxis, angioedema, drug fever, erythema multiforme, generalized skin eruptions, urticaria, photosensitivity, rash, pruritus (itching), serum sickness–like syndrome, vasculitis, and lupus-like reactions[1][5][8].
  • Gastrointestinal: abdominal pain, anorexia, diarrhoea, elevated liver enzymes or bilirubin, vomiting, hepatitis, pancreatitis, pseudomembranous enterocolitis, stomatitis[1][3][5].
  • Kidney and urinary: increased blood urea nitrogen (BUN) or serum creatinine, crystalluria, nephrotoxicity, interstitial nephritis, renal failure, toxic nephrosis, difficulty or changes in urination, blood in urine[1][5][7].
  • Metabolic: hyperkalemia (high potassium)[1].
  • Musculoskeletal: muscle or joint pain (arthralgia/myalgia), rhabdomyolysis (muscle breakdown)[1][5][7].
  • Respiratory: new or worsening cough, shortness of breath, pulmonary infiltrates[1][7].

Severe skin reactions and blood disorders are rare but require urgent medical attention. Patients are advised to contact a doctor immediately if they develop skin rash, severe weakness, persistent fever, unusual bruising or bleeding, yellowing of the skin or eyes, or signs of an allergic reaction, such as swelling of the face or throat[3][7].
